Accurate pH Measurement after SIP & CIP cycles

Continuous Liquid Chromatography Systems are designed for continuous separation and purification of bio-engineered products like proteins, antibodies, DNA and viruses. pH measurement plays an important role in downstream processing of these products. Controlling the pH of the mobile phase of the chromatograph to the value determined during method development will change the ionization state of the analyte, strengthening the interaction between the analyte and stationary phase and leading to enhanced retention and selectivity. Extremes in pH of the mobile phase can cause degradation of the stationary phase and chromatograph column. The system’s wetted components must be able to withstand clean-in-place (CIP) and steam-in-place (SIP) cycles, which can stress a pH sensor with high heat and caustic. The Rosemount Hx338+ pH Sensor features a pressurized reference junction that works to maintain a drift-free pH signal and fight poisoning ions, even after numerous sterilization cycles.
